Synaptic plasticity in the anterior piriform cortex \(\textit {in vitro}\)
- As part of the olfactory system, the anterior piriform cortex (aPC) is relevant for odorant information processing and odour memory formation. In the present thesis, the occurrence of synaptic plasticity at aPC synapses and the underlying mechanisms were examined in the acute mouse brain slice preparation. Investigations of the frequency dependency of synaptic plasticity at lateral olfactory tract (LOT) and associational (ASSN) aPC synapses revealed distinct plasticity profiles for LOT and ASSN synapses, as well as differences in susceptibility for synaptic potentiation along the longitudinal aPC axis. Studying the contribution of glutamate receptors, the present results show that synaptic potentiation depends on N-methyl-D-aspartate (NMDA) receptors, but is not impaired by specific blockade of GluN2A or GluN2B NMDA receptor subunits. Additionally, metabotropic glutamate 5 receptors play a critical role for the induction and maintenance of synaptic potentiation at aPC synapses.
- Als Teil des olfaktorischen Systems ist der anteriore Piriforme Cortex (aPC) relevant für die Verarbeitung und Speicherung von Geruchsinformationen. In der vorliegenden Arbeit wurde das Auftreten von synaptischer Plastizität und die zugrundeliegenden Mechanismen an aPC Synapsen im Maushirnschnittpräparat untersucht. Experimente zur Frequenzabhängigkeit der synaptischen Plastizität an aPC Synapsen des lateralen olfaktorischen Trakts (LOT) und assoziativer (ASSN) Synapsen identifizierten spezifische Plastizitätsprofile, sowie Unterschiede in der Bereitschaft zur Ausbildung synaptischer Potenzierung entlang der Längsachse des aPC. Die Untersuchung von Glutamatrezeptoren zeigte, dass die synaptische Potenzierung von N-Methyl-D-Aspartat (NMDA)-Rezeptoren abhängt, aber nicht durch spezifische Blockade der GluN2A oder GluN2B Rezeptoruntereinheiten beeinträchtigt wird. Zusätzlich spielen metabotrope Glutamat 5 Rezeptoren eine zentrale Rolle bei der synaptischen Potenzierung an aPC Synapsen.
