Dynamics of the Sodium-D-Glucose co-transporter SGLT1 in mammalian cell lines
Die Dynamik des Natrium-D-Glukose Co-Transporters SGLT1 in Säugetierzelllinien
- The distribution of the Na\(^{+}\)/glucose co-transporter SGLT1 was analyzed by immunocytochemistry. SGLT1 was detected in the plasma membrane as well as in intracellular endosomes associated with microtubules, suggesting dynamics of the intracellular SGLT1 pool. To investigate the properties of SGLT1 dynamics, a GFP-SGLT1 fusion was expressed and analyzed in living COS-7 cells. Analysis of the GFP-SGLT1 fusion revealed that GFP-SGLT1 moves within the cell in a microtubule-dependent manner. The dynamics of GFP-SGLT were further examined at the level of the cell surface. These investigations demonstrated that SGLT1 was constantly appearing in and disappearing from the plasma membrane, thus probably recycling between the plasma membrane and intracellular compartments. The properties of SGLT1 recycling were analyzed and shown to differ significantly from the recycling of the transferrin receptor, suggesting different pathways of the two proteins.
- Die Verteilung des Na\(^{+}\)/Glukosetransporter SGLT1 wurde mit Hilfe immunzytochemischer Methoden analysiert. Dabei wurde SGLT1 sowohl in der Plasmamembran als auch in mikrotubuli-assoziierten Endosomen detektiert. Die intrazelluläre Bewegung von SGLT1 wurde mit Hilfe einer Fusion aus GFP und SGLT1 in lebenden Zellen analysiert. Es konnte gezeigt werden, dass sich GFP-SGLT1 in Abhängigkeit von intakten Mikrotubuli innerhalb der Zelle bewegt. Außerdem wurde die Dynamik von GFP-SGLT1 an der Zelloberfläche untersucht. Dabei konnte die kontinuierliche Bewegung zwischen der Plasmamembran und intrazellulären Kompartimenten, also ein sogenanntes \(\it recycling\) des Transporters, gezeigt werden. Die Eigenschaften dieses \(\it recycling\) von SGLT1 wurden mit dem \(\it recycling\) des Transferrinrezeptors verglichen. Dieser Vergleich ergab, dass die Bewegung von Transferrinrezeptor und SGLT1 unterschiedliche Eigenschaften aufweist und daher entlang verschiedener Wege innerhalb der Zelle abläuft.
