Robust transition metal markers for labelling of peptides via solid phase synthesis methods

  • Several organometallic complexes containing a Mo(\(\eta\)-allyl)(CO)\(_{2}\) or Mo(CO)\(_{3}\) fragment are synthesised and characterised. Subsequently, these are coupled to amino acids and dipeptides and the properties of these conjugates are investigated in detail. Two of these classes of conjugates, namely those based on the complexes Mo(bpa)(CO)\(_{3}\) (bpa = bis-picolylamine) and Mo(His)(\(\eta\)-allyl)(CO)\(_{2}\) (His = histidinate, L or D), display good stability towards dioxygen in solution. These markers are coupled to the N-terminus of leucine-enkephalin (a pentapeptide with primary structure H-Tyr-Gly-Gly-Phe-Leu-OH) via solid phase peptide synthesis methods based on the Fmoc strategy. The conjugates withstand cleavage from the resin and can be easily purified. They were characterised by \(^{1}\)H and \(^{13}\)C NMR spectroscopy and ESI-positive MS and can be detected by infrared spectroscopy. The conjugates with the histidinate complexes were shown to exhibit an interesting partly ordered structure in DMSO-solution.
