Entschlüsselung der enzymatischen Katalyse von kleinen GTPasen durch quantenmechanische und molekularmechanische Simulationen von zeitaufgelösten FTIR-Spektren
- Ziel dieser Dissertation ist es, mit Hilfe von modernen Computersimulationen zu einem detaillierten Verständnis von Reaktionsmechanismen beizutragen, die durch Proteine enzymatisch katalysiert werden. Dazu wurde die Katalyse der Guanosintriphosphat (GTP) Hydrolyse durch das Protein Ras und dessen Effektor-Protein GAP mit quantenmechanischen/molekularmechanischen (QM/MM) Berechnungen simuliert und anschließend mit Ergebnissen aus Fourier Transform Infrarot (FTIR)-Spektroskopie Experimenten verglichen. Ras ist ein zentrales Schalterprotein für das Zellwachstum. In ca. 20% aller Tumore ist Ras onkogen mutiert. Die Informationen aus den experimentell validierten dynamischen Computermodellen geben Aufschluss über die atomar aufgelöste Struktur und die Protein-induzierten Ladungsverschiebungen in den Liganden. Dies führt zu einem detaillierten Verständnis des Katalysemechanismus. Diese Ergebnisse können einen Beitrag zur Entwicklung innovativer molekularer Krebstherapien liefern.
- Goal of this dissertation is a detailed understanding of reaction mechanisms, which are enzymatically catalyzed by proteins. Quantum mechanical/molecular mechanical (QM/MM) simulations are used to investigate the catalysis of the guanosine triphosphate (GTP) hydrolysis by Ras and its effector protein GAP. These theoretical results are compared to experimental data from Fourier transform infrared (FTIR) spectroscopy. Ras is a crucial switch in the signaling cascade for cell growth. In about 20% of all tumors Ras has an oncogenic mutation. The information of the experimentally validated dynamic computer models resolve the atomistic structure and the protein-induced charge shifts of the ligand. This leads to a detailed understanding of the catalysis mechanism. These results could contribute to the development of novel molecular anti cancer therapies.
