Modulation of olfactory signaling
- The survival of most animals is directly associated with the accuracy of their olfactory system. To maintain signal specificity in the olfactory system, many mechanisms have evolved to control signal initiation, duration and termination. Understanding the complex mechanisms underlying these modulation processes requires extensive knowledge about the various molecular components involved. Modulation of olfactory signaling in the periphery can be achieved by regulating the amplification of the olfactory signal, the adaptation of olfactory sensory neurons or by maintaining the olfactory signaling specificity. The present work intends to elucidate (1) the molecular identity of the olfactory calcium-activated chloride channel, (2) the internalization processes of olfactory receptors in native olfactory sensory neurons and (3) the role of the scaffolding protein MUPP1 in the olfactory system.
- Die Wahrnehmung von Düften bildet die Überlebensgrundlage vieler Lebewesen. Im Laufe der Evolution entwickelten sich daher mannigfaltige Mechanismen um die Spezifität der olfaktorischen Signalübermittlung zu gewährleisten. Die Modulation des olfaktorischen Signals in der Peripherie kann erreicht werden durch die Regulation der Amplifikation des Signals, die Adaptation olfaktorischer sensorischer Neurone oder der Aufrechterhaltung der olfaktorischen Signalspezifität. Im Fokus der vorliegenden Arbeit stand daher die Untersuchung molekularer Bestandteile der olfaktorischen Signalkaskade. Dabei wurden vorrangig zwei Aspekte untersucht: die Identifizierung des signalverstärkenden olfaktorischen Kalzium aktivierten Chloridkanals und die Adaptation in olfaktorischen Neuronen durch die Untersuchung der Duftstoff induzierten Internalisierung des olfaktorischen Rezeptors. Des Weiteren gelang es die Rolle des Scaffolding Proteins MUPP1 im olfaktorischen System zu untersuchen.
