Proteomics of membrane proteins
Proteomanalyse von Membranproteinen
- A new approach for the analysis of the membrane proteome of \(\textit {C. glutamicum}\) was developed. The screening for effective pre-fractionation techniques resulted in an optimized high salt wash and the subsequent solubilization of the membrane fraction by the detergent ASB-14. Anion exchange chromatography in combination with SDS-PAGE (2-DC) was optimized for separation. Detailed analysis revealed that this approach can display hydrophobic proteins; 50 out of 170 identified proteins were membrane integral, containing one to 13 TMHs and a GRAVY-score up to 0.8. In parallel 16-BAC/SDS-PAGE was tested for the separation of the membrane proteins but was clearly inferior to the 2-DC approach. The membrane proteome of a variety of mutants and cells at different growth conditions was analyzed. Gels were highly reproducible and applicable for quantification. A whole set of membrane proteins was found to be regulated under the tested conditions and allowed the interpretation of regulation mechanisms.
- Für die Analyse des Membranproteomes von \(\textit {C. glutamicum}\) wurde eine Kombination aus Anionenaustausch-Chromatographie und SDS-PAGE entwickelt (2-DC). Eine optimalen Vorfraktionierung der Membranproteine wurde durch eine Hochsalzwaschung, gefolgt von der Solubilisierung mit ASB-14 erreicht. Analysen demonstrierten die Vorzüge der 2-DC Methode; 50 von 170 gefundenen Proteinen waren membranintegral, sie hatten bis zu 13 Transmembrandomänen und einen GRAVY-Score bis 0,8. Weiterhin wurde die 16-BAC/SDS-PAGE für das Membranproteom von \(\textit {C. glutamicum}\) optimiert. Für diese Methode zeigten sich allerdings deutliche Limitationen. Das \(\textit {C. glutamicum}\) Membranproteom wurde unter dem Einfluss verschiedener Parameter untersucht. Die Gele waren hoch-reproduzierbar und für quantitative Analysen mittels Densitometrie geeignet. Die Identifizierung regulierter Membranproteine erlaubte die Interpretation der Daten in Bezug auf verschiedene Regulationsmechanismen.
