A contribution to the understanding of molecular process of biomineralization
- Apatite is an important biomineral, which constitutes the endoskeleton (bones and teeth) in vertebrates including humans. The interaction between the apatite surface and biological environment plays a key role in biomineralization. To understand the interaction between apatite and organic species requires atomic scale information of the apatite surface and the apatite-biomolecules interface. In the work, the structure of fluorapatite (100) surface was first investigated in dry environmental conditions using Grazing Incidence X-ray Diffraction (GIXRD). To understand the apatite-biomolecule interface, experiments were performed under humid and completely hydrated conditions, which were then followed by apatite-aqueous glycine interface study. Measurement and analyses of specular and non-specular crystal truncation rods in all cases provided information about the surface structure of mineral and sites of adsorption on atomic scale. Simulation studies supported the experimental results.
- Apatit ist ein wichtiges Biomineral und Haupt-Bestandteil des Endoskeletts von Wirbeltieren, z.B. Menschen. Die Wechselwirkung zwischen Apatit-Oberfläche und der biologischen Umgebung spielt eine entscheidende Rolle in der Biomineralisation. Zum Verständnis der Wechselwirkung zwischen Apatit und organischen Molekülen, sind Strukturinformationen auf atomarer Skala von der Apatit Oberfläche und der Apatit-Biomolekül Grenzfläche erforderlich. Es wurde die Struktur der (100)-Fläche von Fluorapatit in trockenen Umgebungsbedingungen mithilfe von Röntgenbeugung unter streifendem Einfall untersucht und Experimente unter humiden Bedingungen bis hin zur vollständig hydratisierten Oberfläche durchgeführt, gefolgt von der Untersuchung der Apatit-Grenzfläche in Kontakt mit einer wässrigen Glycin-Lösung. Die Analyse von Crystal Truncation Rods lieferte Informationen über die atomare Oberflächenstruktur und Adsorptionsplätze von Molekülen. Simulationen bestätigten die experimentellen Ergebnisse.
