Kristallographische und biochemische Untersuchungen zum Proteinkomplex Ran·RanBP1·RanGAP
- Die hier vorgestellte Kristallstruktur des ternären Komplexes Ran·RanBP1·RanGAP zeigt das Guaninnukleotid-bindende Protein (GNBP) Ran im Komplex mit RanBP1 und dem GTPase-aktivierenden Protein (GAP) RanGAP. RanGAP besitzt keinen "Arginin-Finger", für den bisher eine universelle Bedeutung für die GAP-stimulierte GTP-Hydrolyse postuliert worden war.
Die kinetische Analyse von Proteinmutanten nach Michaelis-Menten stellt heraus, dass die Ausrichtung des Glutamin 69 durch GAP für die 100000-fache Stimulation essentiell ist. Dadurch kann Glutamin 69 das Wassermolekül für den nukleophilen Angriff am Phosphat optimal positionieren.
Die kinetische Analyse der Protein-Protein-Interaktionen ergibt, dass RanBP1 nur eine untergeordnete Rolle bei der GTP-Hydrolyse spielt, dass RanGAP und Ran auf einer sehr schnellen Zeitskala interagieren und dass die saure, C-terminale Region von RanGAP nicht für die Bindung an Ran, sondern für die korrekte Ausbildung des mitotischen Spindelapparates bedeutsam ist.
- The crystal structure of Ran·RanBP1·RanGAP presented here shows the guanine-nucleotide binding protein (GNBP) Ran in complex with RanBP1 and the GTPase-activating protein (GAP) RanGAP. RanGAP does not employ an "Arginine-finger", which had been assumed to be crucial for GAP-stimulated GTP hydrolysis.
The kinetic analysis of protein mutants based on the Michaelis-Menten formalism shows that the alignment of Glutamine 69 by GAP is crucial for the 100000-fold stimulation. By an optimal alignment of Glutamine 69 can position the water molecule for the nucleophilic attack on phosphate.
The kinetic analysis of protein-protein-interactions shows, that RanBP1 is only of small importance for GTP hydrolysis, that RanGAP and Ran interact on a very fast timescale, and that the acidic, C-terminal region of RanGAP is not important for binding to Ran, but for the correct assembly of the mitotic spindle apparatus.
