Examination of angiotensin II mediated signaling pathways in podocytes via mass spectrometry
- The Angiotensin Type 1 receptor (AT1R) signaling pathway was analyzed in several cell lines after Angiotensin II (Ang II) stimulation via mass spectrometry. This study aims for the investigation of the highly complex interactions within this signaling pathway. Its comprehension within podocyte cells is of interest to find specific approaches to successfully treat various diseases afflicting the kidney. By using SILAC combined with phosphopeptide enrichment and high resolution mass spectrometry it was possible to identify putative novel proteins in the AT1R signaling network. A total of 306 phosphoproteins with regulation was obtained across three different data sets collected from HEK293T, undifferentiated and differentiated AB8 cells. Most notably, the phosphoproteom of the differentiated AB8 cells was in focus for this study, amounting to 139 regulated phosphoproteins. Furthermore, the attempt was made to categorize the identified proteins into broader fields of molecular function.
- Der Angiotensin Typ 1 Rezeptor (AT1R)-Signalweg wurde in diversen Zelllinien nach Angiotensin II (Ang II)-Stimulation per Massenspektrometrie untersucht. Die Studie erforscht die höchst komplexen Interaktionen innerhalb dieses Signalweges. Dieses Verständnis innerhalb von Podozytenzellen könnte dabei helfen spezifische Behandlungsmethoden für unterschiedliche Nierenkrankheiten zu entdecken. Mit Hilfe von SILAC, der Anreicherung von Phosphopeptiden und Massenspektrometrie war es möglich Proteine zu identifizieren, welche potentiell am AT1R-Signalweg mitwirken. Insgesamt wurden 306 Phosphoproteine mit Regulation in HEK293T, undifferenzierten und differenzierten AB8 Zellen festgestellt. Von besonderem Interesse waren dabei differenzierte AB8 Zellen, welche 139 regulierte Phosphoproteine aufwiesen. Des Weiteren wurden die identifizierten Proteine in Felder nach ihrer molekularen Funktion eingeordnet.
