Towards the regulation of ASK-1 by ras activity in the mouse brain
- Counteracting signalling pathways are frequently part of a mutually regulated network allowing the fine-tuned cellular response to diverse stimuli. This thesis deals with the elucidation of a possible co-regulation of HRas and ASK-1 mediated signalling pathways. HRas is a monomeric GTPase controlling survival, growth and other essential processes, whereas ASK-1 is mostly studied in the context of apoptosis. Overexpression of constitutively active (G12V) HRas leads to a diminished ubiquitination and degradation of ASK-1 after stress. However, ASK-1 downstream effector activity is modulated by G12V Ras, too and the pattern of ASK-1 interacting proteins is altered under UV-stressed conditions. Apart from its role in apoptosis, ASK-1 kinase activity stimulates dendritic elongation in hippocampal mouse neurons. This effect is even more pronounced in cells overexpressing activated Ras.
- Gegenläufig regulierte Signalwege sind häufig ein essentieller Bestandteil zellulärer Signaltransduktion und dienen vor allem der Feinabstimmung der Zellantwort auf unterschiedlichste Reize. Diese Arbeit beschäftigt sich mit der Untersuchung einer potenziellen Ko-Regulation von HRas und ASK-1 gesteuerten Signalwegen. HRas ist eine kleine GTPase, die vor allem für ihre Steuerungsfunktion von Zellüberleben und Wachstum bekannt ist, ASK-1 hingegen ist essentiell für die Kontrolle des kontrollierten Zelltods (Apoptose). Die Überexpression von konstitutiv aktivem (G12V) HRas bewirkt eine verringerte Degradierung von ASK-1 nach UV-Stress; außerdem moduliert HRas die Aktivität der Signalgebung von ASK-1, wohingegen UV-Stress zu einer Veränderung des ASK-1 Interaktoms führt. Darüber hinaus stimuliert ASK-1 Aktivität das Auswachsen von Dendriten in hippocampalen Neuronen der Maus. Dieser Effekt ist besonders stark in Zellen, die konstitutiv aktives HRas über-exprimieren.
